Microarray
In 1999, the YCC/Keck Microarray Shared Resource (MSR) was established with support from the YCC, Yale School of Medicine, Departments of Genetics and Pathology, and the Keck Laboratory. After its inception, it rapidly began to provide services to support a wide spectrum of cancer and biomedical research at Yale and many other Universities.
Specific Services
The Affymetrix section of the MSR is a full service facility that offers services, reagents, equipment, and expertise required for perfoming large-scale gene expression studies utilizing commercially available oligonucleotide based Affymetrix microarrays. Major Affymetrix services provided include the assistance in planning and carrying out microarray experiments; expression analysis services that include RNA/mRNA extraction, preparation of target samples suitable for array hybridization, and data analysis; SNP analysis using both Affymetrix (10,100 and 500 K assays) and ABI platforms; and quantitative PCR validation of microarray results using the ABI 7900 Sequence Detection System. The MSR also offers a range of spotted glass slide microarray services with investigators having the option to use as few or as many services as are needed.
There are a number of glass slide services available:
- Array on to slides standardized human, mouse, rat and other oligonucleotide and cDNA sets from commercial and academic sources.
- Array custom nucleic acids (PCR products, oligonucleotides, cDNAs) and proteins/antibodies from individual investigators.
- Provide pilot and complete project services using a variety of dye labeling approaches, dependent upon RNA quantity, array type and application.
- Hybridize custom fluorescently labeled probes, scan the resulting slides, and acquire image and numerical data.
- Analyze the resulting data to generate expression ratios, scatter plots, cluster analyses etc.
- Provide consultation for experimental design, grant writing and project development.
- Provide bench training in micrarray protocols and analysis methods.
- Assist investigators in transferring data to public repositories via Yale Microarray Database (YMD) before publication.
